Plumbing a new radiator question

My credo
We need to stop messing with the framework that the founding fathers created.
I’m changing out my radiator today and have a question about how to seal the trans cooler lines. Do I need to use any sort of thread sealer or just tighten it until it doesn’t leak?

Any other hints about bleeding air out would be welcome too.

How about lower rad hose... doesn’t have to be the molded one or can I us there flexible one?
 
Transmission lines are typically double flared and require no sealant. They seal by the two tapers coming together.... >>

Lower radiator hose is the suction side for the water pump. They typically have an internal spring to prevent the hose from collapsing under suction. I believe most flex hoses will have enough rigidity to prevent collapse

Typically when filling that type of cooling system I will fill the radiator, then pop a single heater hose off, stick a funnel in it, and fill the hose until coolant comes out of the heater core side. This removes most of the air from the block and prevents an air-bound core by filling from the highest point.
