Misfire cylinder #6 next steps

Hey guys my truck has idled rough from time to time and recently it was idling really rough at a stoplight and the check engine light popped on. I pulled the code and it was a misfire on cylinder #6. I reset the computer and replaced my plugs and wires (needed it either way pretty sure they were all originals on a 2000 and the gaps on the plugs were way out of spec). That has not seemed to remedy the problem, it still idles rough from time to when when stopped and in gear. However once i give it some gas it runs like a top and then usually will not repeat the problem even at the next stop sign/light. Any thoughts of what I should be checking out next would be much appreciated!
 
Can you switch switch out a coil pack to see if the problem moves? Compression test (hot engine). How did the old #6 plug look? Switch injector to see if problem moves.
