The transfer case has nothing to do with the rear differential and is totally seperate. I think you're referring to the cover on the diff? If so, yes you have to remove it because there is no drain plug. The fluid spills out when you take the cover off. And it is a good idea to clean it out while it's off. As for fluid, any 80w-90 or 75w-90 fluid would work fine. Synthetic lasts longer and seems to make it smoother, in my experience. But it's much more expensive than conventional.
