"Manual" door lock adjustment.

My ranger is a 1986 ext cab. It has manual door locks. I replaced the door due to some dumbass t boning my truck when it was parked. I bolted the handle back on the door I got from pick and pull. The screws would come loose, so when I was working on my truck last weekend I put some locktite on the screws holding the handle on. Of course later in the day I went to lock my truck and it would not lock. Anyone have any tips to adjusting the lock assembly?
 
Does it lock with the key or the interior button?

It's really hard to say without seeing it, but I would suggest removing the door panel and checking to see if the rods are attached properly, the nut on the lock cylinder is tight, the cylinder is installed the right way.

Does the rod have one threaded end? if so than you can adjust it by opening the clip that holds that end and adjust it either way. If not and all else is good, it may be a problem with the latch mechanism itself and you may have to change it (I'm assuming since you got the door from the pick and pill, you should have another latch)
