Yes, it is possible to convert a 2wd transmission to 4x4. You need the output shaft and tail housing from a 4x4 transmission and to partially disassemble the 2wd transmission and install the 4x4 transmission parts so usually it's easier to just find a 4x4 transmission to start with. You could also use a divorced transfer case with a 2wd transmission but these aren't very common so once again it's easier to use a 4x4 transmission and transfer case in most situations.
